I have Exchange 2010 on Win2008 and BES express on Windows 2008. The mx is ready, BES express is installed and configured. It has live IP and host record. I have 50 BB users.
Now i want to put this BES Express into production, but i am not sure where to start. Should i have to contact RIM for enabling BES Express activation.

There might be several misunderstandings here.
Could you please first clarify whether you are talking about BIS (BlackBerry Internet Service) or BES (BlackBerry Enterprise Server)?
I cannot help you with BIS.
But for BES you might be on the wrong path.
While installing the BESx, you should entered the SRP-ID and Authentication Keys and all, you have been sent by BlackBerry to activate your BESx. So as soon as your BESx is running it should also be connected to (and activated with) the BlackBerry infrastructure. You don't need an MX.
Please check the following:
If all is OK you should be able to activate your devices.

BES express uses BIS dataplans in most countries.
If you want OTA activations you need an MX record so emails can be sent from the BlackBerry NOC when device activates
