A few things to check:
Make sure that your Desktop Manager software is up to date. If you upgraded from an older model, you must upgrade DM for it to work.
Try changing the order of which you do first - connect the cable or start DM, I've found some PC's are fussy about the order.
You may need to uninstall/reinstall your PC's USB drivers and make sure they are up to date.
If you have other USB items plugged in, try removing them temporarily and try different ports.
