The question is not clear to me.
A protocol is a way of interpreting the bytes sent. The underlying protocol for all the data connections is TCP. You can use HTTP protocol (whihc builds on top of TCP) on any of MDS, BIS-B, direct or WAP.
You mention a URL. Most commonly these are associated with the HTTP protocol. So you can access a URL using any of the connection methods mentioned.

i am using the following code in my application
HttpPConnection conn = (HttpConnection)Connector.open(url);
and trying to acces it through wifi ,
in which format the connection will be framed, either TCP , MDS or BIS

I think you need to become more familiar with the various connection options available on Blackberry and how they are specified.
