I've created the software configuration, created the BlackBerry Device Software configuration, assigned the BlackBerry Device Software bundle, and applied the software configuration to the BES User account. The update does not seem to get pushed to the device. How can I manually specify when to push the update and then view the deployment status?

Are you talking about an application update or about an update for the device firmware? The later one only works, if your carrier, from where you bought the devices, supports over the air software load. Currently the number of carriers supporting OTASL is very small, e.g. Vodafone, Verizon, AT&T. For OTASL to work, your devices need to be at least at firmware version 4.5 or newer.
