Hi and welcome to the forums,
I would STONGLY advise against downloading the media manage. If you do a search in the grey box in the upper left hand corner, you will notice that the MM is not stable. I find that if you have a media card, the best way is to connect the BB and use drag and drop.
Make sure there folders are on the media card,
This is where the Blackberry will look for the associated files.
Bit I got ahed of my self, DO you have a media card?
Please let me know!

Hi and thanks for the fast response. I need to clarify the issue. The Media Manager was at 1 time able to connect to the device. I used the media manager to successfully copy a ring tone from my PC to the Blackberry. I then had encountered had some Lotus Notes issues on the handheld and had to wipe the unit and reactivate using Enterprise Activation. All was well until recently. Now the Roxio Media Manager just will not connect to the device.
I do not have a media card but that will change tonight. When the card is installed how would I then transfer ring tones and other stuff to the device without using the Media Manager

Go to options media card and enable all options except encryption mode set to none.
Make sure the following directories are listed on your media card,
In lieu of media manager which is unstable. I would drag and drop the files to the media card using my computer.
Please let me know your thoughts!

I inserted the card and found that there was a Blackberry folder containing the other folders already coded. My guess is that the 8330 does that by itself. I copied a mp3 file over as well as a ring tone. When I copied the ring tone a Roxio message dialog popped up informing me that some of the data would not be copied. I copied the file anyway and took note that the sound quality had degraded a bit. It is my understanding that 1 of the things that the Roxio Media Manager tries to do is further compress both ring tones and mp3 files. However, everything appears to be working fine.
It's a shame that the Roxio portion of the Desktop Manager doesn't function. It seems that Rim should be working with them to patch the their program.
Thanks much for all your help and support.

If you have read as may posts as I have, you would never hd installed it. It's very unstable and adds another fail point to your PC.
most users use the drag and drop method.
Glad you have it working.
Make sure you have these options set in STATUS MEDIA:
Media Card window
Mass storage support
auto enable when connected
If you need further assitance please ask! If not:

I have stopped using and worrying about the Media Manager.
The only option not correctly set was "Auto Enable ... when connected". I have set this option to Yes.
Thanks again for all you help and advice.
